چکیده
J. TECHNIC was (lescribe(l by Few (1) in 1961 for deterniiiiation of I 7-hydroxycorticosteroids in which borohydride re(luction of steroid 20-keto groups was followed by oxidation of adjacent hydroxyl groups in the 17-20 positions with nletaperio(late. The borohydride also reduced all preformed 17-ketosteroids to the corresponding alcoholswhich do not give the Zimmerman reaction-alici reduced steroid 11 keto groups to hydroxyl groups. The metaperiodate also oxidized steroid glucuronides to formates, which are easily hydrolyzed by alkali. These reactions are shown in Fig. 1. The 17-ketosteroids formed were separated by partition chromnatography and determined by means of the Zimmerman reaction, using appropriate standards. The only ketosteroids which were found to be invariably preseiit were etiocholanolone and ii -hydroxyetiocholanolone (1). It has been found that these can be separated adequately by simple solvent partition without nee(l for a chromatographic step.

A note on the rapid assay of 17-ketogenic steroids in urine.
The determination of urinary corticosteroids by their conversion to 17-ketosteroids (17-KS) on treatment of urine with sodium bismuthate has been recently reported (Norymberski, 1952; Norymberski, Stubbs, and West, 1953). The group of corticosteroids thus determined has been termed 17-ketogenic steroids (17-KGS). The interest in this method shown by colleagues from other laboratories prompts us...
متن کاملA procedure for the measurement of urinary 17-hydroxycorticosteroids, aldosterone acid-labile conjugate, and free plasma cortisol in patients with chronic illness.
The usualstepsfor steroid isolationare: (1) hydrolysis, acid for the urinary acid-labile aldosterone conjugate, and enzymatic for the urinary 17-hydroxycorticosteroids (17-OHCS); (2) organic solvent extraction of the corticosteroids; and (3) alkaline, acid, and aqueouswashing of the organic extracts of steroids. In the method described the crude extracts are purified by 3 or 4 sequential partiti...
متن کاملTHE DETERMINATION OF URINARY 17-TRIOXYSTEROIDS* BY RAYMOND RIVOIRE,t
The color reaction characteristic of cortisone and related compounds (with the 17,21-dihydroxy-20-keto grouping), which Porter and Silber (1) reported in 1950, has been used by a number of investigators to measure the “cortisone-like” metabolites of urine. To facilitate expression, we propose to give to the methods of analysis utilizing this reaction the term of “determination of 17-trioxystero...
متن کاملA case of Cushing's syndrome with pigmentation and severe hypokalaemic alkalosis.
Hypokalaemia is the most important result of the adrenal hyperfunction seen in the recently described Conn's syndrome (Conn and Louis, 1956). In this syndrome aldosterone is produced excessively, but evidence of the same excess of other hormones is lacking and the excretion of 17-ketosteroids and 17-ketogenic steroids in the urine is normal. Hypokalaemia may also occur in cases with a high urin...
متن کاملClinical Value of Urinary 17-ketogenic Steroid Determinations
During recent years increasing importance has been attached to the study of adrenal cortical hormones, and consequently much work has been directed towards utilizing the daily output of these hormones and their metabolites in urine as an index of adrenocortical function. Hydrolysis of the conjugated forms in which the steroids are present in urine is a necessary preliminary to their extraction ...
